Disruption of Circadian Feeding Patterns in Obese ZDF Rats

Abstract Body

Background: In humans, shift work and the resultant circadian misalignment in eating patterns are associated with an increase in metabolic disorders, including type 2 diabetes mellitus and obesity. Our aim was to see if circadian eating patterns in obese Zucker Diabetic Fatty (ZDF) rats are disrupted compared to lean littermates.

Methods: We assessed food intake, weight and serum glucose in ZDF rats (n=18) and lean controls (n=18). Food consumption was monitored between light and dark to look for disruption of normal circadian patterns. Groups were compared using t-test.

Results: ZDF rats consumed a higher percentage of food during light hours compared to lean littermates (43.6% vs. 36.7%, respectively, p<0.01). Total daily food consumption was greater in ZDF rats compared to lean controls even when adjusted for weight (53.4mg vs. 32.2mg per gram body weight per day, respectively, p<0.01). Despite increased intake, weight gain was less in ZDF rats. ZDF rats had higher serum glucose levels too.

Conclusion: In ZDF rats the regular circadian pattern of food intake is disrupted, with increased total and weight adjusted food intake for this group, and an increase in serum glucose. Despite increased food intake, the weight increase of the ZDF animals was less than lean controls. This paradoxical reduction in rate of weight gain despite increased intake, and possible relation to the disrupted circadian food intake needs further investigation.
